Supercomputers being harnessed in the battle to block cancer-causing proteins

As the quest for drugs to combat ever more specific diseases gets ever more involved, so the human brain often requires extra help in coming up with solutions. Scientists at Ohio State University have decided, therefore, to arm themselves with what could be considered the ultimate weapon in the battle for long-lasting and effective medicines – a supercomputer.

They are hoping that the computer’s massive processing power will enable them to block small molecules of the protein Interleukin-6, which is a vital messenger which the body sends when it needs to combat traumas, burns or infections. In people with cancer, Interleukin-6, or IL-6, is produced to excess, and therefore needs a signal in order to turn it off once it has produced the right level of the protein.

Some years ago, Japanese scientists believed they had found a means of reducing the signal produced by IL-6 to help them achieve this artificially. As is often the case, commercial reconstruction of this process has proved prohibitively expensive, but the biggest setback to the researchers came when they discovered that the marine bacteria which they used to suppress the IL-6 naturally mutated to produce a compound which was far less effective.

The professor behind the research, Chenglong Li, remains, however, hopeful that by harnessing the joint skills of the university and the neighbouring James Cancer Hospital, the compounds which were found to have the powerful healing properties can be recreated in a new, and possibly even more potent, form.

And early research suggested that this could be the case. Using the advanced molecular modelling capabilities of the Ohio State University’s advanced computers and modelling simulation software, he has been able to observe how these proteins interact, and find the best spot for attaching the two ‘helper proteins’ which carry the signal needed to turn off the production of IL-6.

They key to success in this research lies in creating bonds which are strong enough to produce protein-blockers with the highest level of effectiveness, has been the microscopic examination of more than 6,000 drug fragments. The ultimate aim is to produce a molecule with powerful bonding capabilities, because this will be essential in making the original molecules easier to produce in synthetic form as an ingredient for an eventual drug to treat individual cancers – specifically breast tumours.

Such computational capability, while holding the key to this particular research, also looks likely to play a vital role in other heavily-funded drug development programmes for tackling other cancers, such as prostate and ovarian. And with massive US government funding behind them, there is a great deal of hope that Dr Li’s computational chemistry techniques might prove crucial in making significant breakthroughs in this field.

It is unsurprising that various cancers are the focus of intensive activity in
current medicinal chemistry
. The use of ‘super-technologies’ in
drug development
must be considered a vital step to helping take human research so much further in the quest for cures.
